Cancer invasiveness and metastasis is still a mystery. With growing cancer incidence the need of understanding tumor cell migration in its complex microenvironment provide an important impact for cancer therapy. Fibroblasts play a key role in the malignant progression of cancer and are therefore an important target for cancer therapies. The development of miniaturized 3D in vitro models for the investigation of cancer cell migration could contribute to a better understanding of the fibroblast role and therefore the development of new anticancer agents.
At the LCMD we developed a method to produce millimeter-sized spherical core-shell structures using biopolymers as the shell material named “liquid pearls”. In cooperation with the Institut Curie we already use the liquid pearls for tumor spheroid production. This method is planned to be adapted and combined with specific developed microfluidic PDMS gradient devices which enable the investigation of cancer migration through an extracellular membrane (EM) after the breakthrough of the primary solid tumor through the basal membrane.
The aim of the master project is to modify and optimize the existing PDMS gradient device in terms of

1. Stable, longterm gradients using fluoroprobes and fluorolabelled chemoattractants,
2. Characterization of the gradients and determination of diffusion coefficients,
3. Spheroid placement in the collagen channels and
4. First cell migration studies.

In this work an interface between material science, biophysics and microfluidic has to be established and a strong interest working in an interdisciplinary field is a requirement. We are searching for ambitious students which like to work interdisciplinary within an international group with a strong interest in cancer research field. This six months project will take place in the LCMD laboratory at ESPCI under supervisory control of Dr Anette Funfak and Pr. Jérôme Bibette.

Des bactéries sont capables de métaboliser des hydrocarbures en formant des biofilms à l interface eau/huile, lui conférant ainsi un caractère viscoélastique. De plus, certaines de ces bactéries sécrètent des agents de surface qui affectent aussi les propriétés de l interface eau/hydrocarbure.
Pour ce stage, nous proposons d élaborer une méthode de tri de bactéries, pouvant métaboliser des hydrocarbures, à l aide d un dispositif microfluidique. L idée est d encapsuler différentes souches de bactéries dans des gouttes d émulsion eau dans l huile et de tirer partie de l abaissement de la tension de surface générée par les bactéries d intérêt afin de les sélectionner. Une étude préalable sera réalisée sur la mesure de l évolution de la tension de surface en présence d une souche métabolisant des hydrocarbures. Un dispositif microfluidique sera mis en place pour permettre l encapsulation de bactéries dans des gouttes d émulsion suivie de leur incubation et enfin d un tri hydrodynamique des gouttes, et donc des bactéries, d intérêt. Une démonstration du principe sera conduite à partir de deux souches de bactéries dont une seule est capable de dégrader des hydrocarbures.

Lorsque la taille d un système diminue, ses fluctuations augmentent: ainsi, pour une enzyme ou une particule colloïdale, les variables thermodynamiques comme le travail ou la chaleur ne sont définies que de façon statistique. Pour ces raisons, ces systèmes sont décrits par la thermodynamique stochastique. Les distributions statistiques associées aux variables thermodynamiques obéissent à des relations très générales, appelées relations de fluctuations, qui sont au centre de développements récents dans le domaine de la Physique Statistique des systèmes hors de l équilibre. Afin d étudier ces questions sur un exemple simple, une expérience modèle a été réalisée au laboratoire. Elle consiste à manipuler deux particules colloïdales magnétiques, dont la position peut être mesurée en fonction du temps par vidéo-microscopie. En appliquant un champ magnétique variable dans le temps, on peut suivre la dynamique de ces particules colloïdales en présence du champ et quantifier les distributions de chaleur et de travail correspondantes. Au cours de ce stage, on cherchera à mieux comprendre les relations de fluctuations vérifiées par ces distributions, et à utiliser ces relations pour caractériser la dissipation présente dans ce système modèle hors d équilibre. Ce travail se fera en collaboration sur la partie théorique avec  D. Lacoste au laboratoire Gulliver de l ESPCI.

Position description: Postdoc position for improving a droplet-based millifluidic platform for microbial diagnostics and validation on clinical cases.
Industrial partnership: ESPCI (http://www.lcmd.espci.fr) and bioMérieux (http://www.biomerieux.com), the world leader in bacterial diagnostics.
Scope of the project: Bacterial infections are a major health problem concerning worldwide more than 1.3 millions people everyday. Appropriate treatments rely strongly on the quality of the diagnostics, that is, the identification of the pathogens and of their resistance profiles. The antibiogram is one of the tools that enables clinicians to determine resistance to antibiotics and thus guides them in their decision-making. The increase in resistant bacteria and in the number of antibiotics necessitates the use of high-throughput solutions. The Laboratoire de Colloïdes et Matériaux Divisés at ESPCI has developped millifluidic tools for the control of droplets dispersed in an oil phase (water/oil emulsion) within tubing. Each droplet is used as an independent microreactor. By mixing a bacterial suspension and changing antibiotics concentration within every droplet we demonstrated the measurement of high resolution antibiograms.
Goals of the project :The long term objective is to develop an instrument for antibiograms determination that would be superior in term of quality of results and throughput than the gold standards.
To reach this goal we first have to :
1. improve the performances and integration of the existing technology
2. validate its universality across a large spectrum of clinical cases
The first half of project will be dedicated to technological development and the second half to biological studies in strong collaboration with experts from bioMérieux.

Start date and duration of contract: The expected start date is 1^st March 2013. The salary will be up to 2,500 Eur/month depending on experience. The project will be for 1 year.
